Polylactic Acid and Polycaprolactone Blended Cosmetic Microneedle for Transdermal Hispidin Delivery System

Microneedles (MNs) are a new system of effective drug delivery that create micron-sized pathways to the epidermis or upper dermis regions skin. In this study, we developed coated-type microneedles for direct hispidin Hispidin is well-known plant-derived antioxidant component showing antitumor, anti-inflammatory, antiallergic, antiangiogenic, antioxidant, hypoglycemic, hypolipidemic, and immunomodulatory activities. Polymeric blends polylactic acid (PLA) polycaprolactone (PCL) were casted as MNs enhance skin permeability. PLA/PCL exhibited highest strength 51.26 MPa with width ~200 ųm. was directly coated onto form layers. Compared hispidin-only layer, permeability increased by over 50% when using agarose gel in vitro tests. dose-dependent manner, on also showed brightening effect, well anti-inflammatory activity at gene protein level cell culture experiments. It demonstrated antimicrobial activity, no cytotoxicity cells. These results suggest MN may have great potential prototype platform various systems, allowing development more subcutaneous vaccines, oligonucleotides, insulin, many other cosmetic applications.
